Comparison of Carbonylation in Young and Old Rat Skeletal Muscle

چکیده
Since muscles support almost all the activity of our bodies, the relationship between aging and muscle capability is important. Dr. Edgar Arriaga from the Department of Chemistry studies the effect of age on muscle damage for different muscle types and cell regions. As part of the summer statistical collaboration program, I was on a team that collaborated with them to analyze data from a recent experiment. We built models for two cell regions separately, and also combined them into a larger model. Models included several levels of random effects. We found no evidence of age effect or muscle type effect, but did find evidence of region effect. Results are shown both numerically and graphically. Background Dr. Edgar Arriaga from the Department of Chemistry studies the effect of age on muscle damage for different muscle types and cell regions. His lab had undertaken a study to characterize oxidative damage in aging skeletal muscles. Oxidative damage is a particular kind of muscle damage, while age, muscle type and cell region are the potential effects to the damage. So they chose samples with different ages and different muscle types and measured each sample from both regions. The researchers chose rats for this experiment, due to low cost and muscle similarity between rats and human.

Protein Carbonylation and Heat Shock Proteins in Human Skeletal Muscle: Relationships to Age and Sarcopenia
Aging is associated with a gradual loss of muscle mass termed sarcopenia, which has significant impact on quality-of-life. Because oxidative stress is proposed to negatively impact upon musculoskeletal aging, we investigated links between human aging and markers of oxidative stress, and relationships to muscle mass and strength in young and old nonsarcopenic and sarcopenic adults. Sixteen young...
متن کاملThe Effect of Low Volume High Intensity Interval Training on Sarcolemmal Content of Fatty Acid Transport Proteins (FAT/CD36 and FABPpm) in Young Men
High-intensity interval training (HIT) induces skeletal muscle metabolic and performance adaptations that resemble traditional endurance training despite a low total exercise volume. On the other hand, fatty acid oxidation is increases in skeletal muscle with endurance training. This process is regulated in several sites, including the transport of fatty acids across the plasma membrane. The...
متن کاملAging and acute exercise enhance free radical generation in rat skeletal muscle.
Reactive oxygen species (ROS) are implicated in the mechanism of biological aging and exercise-induced oxidative damage. The present study examined the effect of an acute bout of exercise on intracellular ROS production, lipid and protein peroxidation, and GSH status in the skeletal muscle of young adult (8 mo, n = 24) and old (24 mo, n = 24) female Fischer 344 rats. Young rats ran on a treadmi...

Background and Objectives: Skeletal muscle mass depends on the balance between synthesis and degradation of muscle protein, which changes with aging and disease. The aim of the present reserch was to examine the effects of two exercise training protocols on alterations of some genes involved in pathways of protein synthesis and degradation in order to achieve a more effective training program i...
متن کاملThe superoxide dismutase mimetic tempol does not alleviate glucocorticoid‐mediated rarefaction of rat skeletal muscle capillaries
Sustained elevations in circulating glucocorticoids elicit reductions in skeletal muscle microvascular content, but little is known of the underlying mechanisms. We hypothesized that glucocorticoid-induced oxidative stress contributes to this phenomenon. In rats that were implanted with corticosterone (CORT) or control pellets, CORT caused a significant decrease in muscle glutathione levels and...
